Analysis of Dielectrophoretic Properties of Cells by the use of the Uniform Field Gradient

Popis: Dielectrophoresis (DEP) has been widely used for manipulation, concentration, and discrimination of biological cells. Most of developed DEP devices have been parallel-plate type flow channels. Because the magnitude of the DEP force is proportional to the gradient of field square, the DEP force induced on cells in the coplanar channel takes the maximum at the edges of the electrodes while it decays with height above the electrode at the bottom; enabling a strong non-uniform electric field favorable for cell manipulation. However, there has been less emphasis on the development of analytical DEP devices due to the inherent non-uniform nature of the field. We have developed a device where the gradient of field square is constant; enabling a constant DEP force application to cells in the analysis region in the device. With this approach detailed dielectric measurements of each individual cell is possible with a greater throughput than the current practice using electrorotation. In the present study, the theory and advantages of the proposed device are presented.
