Polypeptide β-sheet detection by cyanine dyes

Popis: Mixtures of oxacarbocyanine or thiacarbocyanine dye and protein- or polypeptides aggregates show a shifted absorption band for the dye compared to its aqueous solution in the case that all or most of the protein- or polypeptide aggregates have β-sheet structure. The shift can be a red or a blue of 10 nm to 25 nm, depending on the kind of cyanine dye. In contrast are mixtures with protein or polypeptides with helical or random coil conformations, where no change in the position of absorption maxima of dye was observed. That is, if possible, also a way to analyze different secondary structures in thin polypeptide and protein adsorption layers and membranes.
