Publisher Summary This chapter describes a system for computer-assisted 3-D reconstruction of serial sections that can be obtained by any laboratory at a relatively low cost. It also discusses technical suggestions regarding specimen handling, data entry, and 3-D image generation that are generally applicable to any 3-D reconstruction system and outlines some basic features of 3-D reconstruction hardware and software that are best suited to the task at hand. The 3-D reconstruction system employed is based on the use of PC3D, a 3-D reconstruction software package marketed by Jandel Scientific. The system can be used across the entire range of magnification, from macroscopic down to the electron microscopic level. PC3D is easy to learn and use, and it provides high-quality, camera-ready 3-D images that are suitable for publication. A set of eight subprograms runs on a standard personal microcomputer. Data are entered as trace points on a digitizing tablet from serial sections that have already been aligned. A 3-D view of the reconstructed object is generated in color and can be displayed from any perspective with hidden lines removed. Analysis of volume can be performed automatically. |
