Popis: | Carbonic anhydrase (CA; EC 4.2.1.1) was purified to electrophoretic homogeneity from spinach leaves. With this enzyme, anti-spinach CA antibody was prepared using rabbits. This antibody precipitated almost all CA activity from a crude extract of spinach leaves. Various immunochemical determinations indicated that the CA protein content in spinach leaves was 1–2% of the total soluble leaf protein. Ouchterlony double immunodiffusion analysis indicated cross-reactivity with leaf extracts from several terrestrial C3 plants, C4 dicotyledons and Crassulacean acid metabolism (CAM) dicotyledons, while the extracts from several C4 monocytyledons and the green alga, Chlamydomonas reinhardtii, showed no cross-reactivity with the anti-spinach leaf CA antibody. |
