Analytical study of extracts of St John’s wort (Hypericum perforatum), evaluation of HPTLC plates by multivariate data analysis

Popis: HPTLC with multivariate data analysis has been used as a new means of evaluating the quality of herbal extracts. Spectral data from three-dimensional chromatograms obtained from St John’s wort ( Hypericum perforatum ) extracts were correlated with opioid receptor affinity to predict pharmacological activity. A total of 27 extracts of St John’s wort from four different accessions extracted with seven different solvents (100, 80, 60, 40 and 20% EtOH in water and 90 and 60% MeOH in water) were analyzed at λ = 200 to 600 nm by use of a diode-array scanner. The correlation between the spectroscopic data and IC 50 values obtained from non-selective opioid binding to rat cortex was determined by means of a quantitative model using a partial least-squares algorithm (PLS-1); the correlation coefficient was 0.97. The model was then verified by prediction of the IC 50 values of seven other extracts of St John’s wort.
