Popis: 1. 1. The cellular uptake of [3H]cortisol has been studied on hepatocytes isolated from the trout Salmo gairdneri irideus. 2. 2. Analysis of the uptake as a function of concentration indicated the presence of a low affinity saturable system (Km=60 μM) and a diffusion component. The saturable part involves metabolic energy and participation of membrane SH-groups. 3. 3. Uptake of [3H]cortisol was decreased when cells were incubated with a large excess of unlabelled cortisol or cortisone given simultaneously. 4. 4. Dexamethasone is much more permeant than cortisol (12 times) and displays an unusual “permissive” effect on cell membrane permeability to steroids. 5. 5. These results suggest that the uptake of cortisol by trout hepatocytes is mediated in part by a carrier protein located in the cellular membrane.
