Popis: Publisher Summary This chapter describes the molecular variation in an intra- and extracellular R-type vitamin B 12 -binding protein. Partially purified LB-B12 and TC I-B12 complexes labeled with different cobalt isotopes were incubated with sialidase for 20 h at 37°C in 0.05 M acetate buffer pH 5.9 containing 0.9% NaCl and 0.1% CaCl 2 . The chapter presents the mobility of sialidase treated TC I and LB compared with samples incubated without sialidase. The sialidase treatment changed the mobility of TC I and LB from α 1 and α 2 respectively to β, but the mobility of TC I was still somewhat higher than that of LB. A part of the LB control underwent the same change in mobility as the sialidase treated sample. The gel filtration of sialidase treated and control-incubated LB showed a slight decrease in size of the treated protein. This difference was very small, and it was demonstrated only when both samples were studied simultaneously labeled with different cobalt isotopes
