Popis: (1) A simple method of demonstrating the cytotoxic effect of diluted tuberculin and PT) on the leucocytes of hypersensitive rabbits is described. The method consists in staining dead and living cells (leucocytes washed four times and suspended in Tyrode solution) with 1% Congo red and 0.2% Nile blue after four hours incubation in vitro. Several hundred experiments were carried out and the results were compared with the state of hyper sensitivity of the rabbits tested. (2) The reaction was negative with the cells of normal rabbits, incubated with and without tuberculin, and with the cells of hypersensitive rabbits incubated without adding tuberculin. It was positive with cells from most BCG-sensitized rabbits mixed with tuberculin. A positive reaction occurred in cases with a high degree of hypersensitivity, while a negative reaction took place in rabbits with low hypersensitivity, as determined by skin tests and the degree of fever in the systemic tuberculin reaction. There was no relationship whatsoever between the titre of haemagglutinating antibodies and the cytotoxic reaction of leucocytes in vitro. (3) A positive reaction (a change in the staining properties of the cells) shows complete correlation with the ability of the cells to release pyrogenic substances on contact with tuberculin in vitro (Johanovsky, 1958).
