Twenty years ago, defective transport of the antimetabolite methotrexate was recognized as a specific cause of resistance to that drug in a mutant line of murine L5178Y leukemia (Fischer 1962). Since that time numerous reports of antitumor drug resistance based on defective drug transport have appeared involving several different groups of antitumor agents. In order to achieve a more complete understanding of this type of drug resistance, the basic mechanisms of drug uptake by mammalian cells will first be reviewed followed by a more detailed description of specific examples of drug resistance due to deficient transport. Next we will consider attempts that have been made to circumvent this type of resistance by increasing drug uptake in resistant transport mutants. The review will conclude with a brief discussion of the possible direction of future studies of this area of investigation. |
