Popis: Publisher Summary This chapter describes results of experiments conducted to study in vitro characterization of the target cell for transfer factor (TF). The experimental team, to test whether the effect of DLE depends on nonadherent cells (T lymphocytes), used the blast transformation assay of Arala–Chaves et al., with adherent and nonadherent cells or nonadherent cells (NAC) alone as the target. Some cultures were supplemented with supernatants from other cell suspensions, to test for possible effects of soluble factors. It is observed that leukocyte suspensions containing roughly 20–30% monocytes and 70–80% lymphocytes were obtained from defibrinated blood by means of centrifugation in a Ficoll–Isopaque gradient according to the method of Boyum. The results reveal that when DLE was tested in cultures containing NAC suspensions in the presence of absence of stimulating antigens, the regression line obtained was linear in both cases with the y -intercept value not significantly different from 1.
