Popis: The organelle composition of plants resulting from protoplast fusion is of great interest in studies on organelle genetics and is very important for plant breeding (1). Ever since protoplast-fusion derived somatic-hybrid plant production became a reproducible procedure the chloroplast composition of the hybrid and cybrid progenies were subjected to analysis (see: 2). The methods applied in these analyses were based on the pigmentation of the plants (e.g. chlorophyll deficiencies), on the expression of the plastome (e.g. streptomycin and tentoxin resistances, electrofocussing of the large sub-unit of ribulose-1, 5 bisphosphate carboxylase) or on the base sequence of the chloroplast DNA (cpDNA) as indicated by the restriction patterns which result from the digestion of cpDNA with endonucleases, followed by gel-fractionation.
