An electrodeless method is described for measuring the intrinsic electron transfer processes that occur in oxidoreductase enzymes. The enzymes may be in free solution, immobilized in membranes, or exist in optically opaque media. Electron injection into the enzyme is accomplished chemically using NAD(P)H. and semiquinone or nitroxide free radicals act as electron acceptors. The rate of electron transfer through the enzyme is determined by monitoring the decay of the radicals using an electron spin resonance spectrometer, and from analyses of the decay kinetics the corresponding logic state diagrams are derived. |
