In order to gain information on the mechanisms which induce a supersensitivity to morphine administration in patients with acute and chronic liver failure, the levels of the endogenous opioid beta-endorphin was studied in plasma and in two brain areas of rats and dogs at two different levels of encephalopathy. In the dog model of portal-systemic encephalopathy the binding characteristics of opiate receptors in hypothalamus and cortex were studied. Herein we report that during the development of encephalopathy there are changes in the levels of beta-endorphins and in the opiate receptor densities which taken overall might explain the altered responce to morphine administration in man. |
