Popis: One aim of our investigations is the optimization of fermentation conditions of hybridoma cells in a fluidized-bed bioreactor. Porous glass carriers (Siran“, Schott) are used for immobilization of the cells. The surface of the glass-carrier are modified with gelatine to reduce the required time for their colonisation. It was demonstrated that this time is reduced twofold with gelatine-modified Siran compared to unmodified Siran. The startup strategy was improved to avoid limiting conditions, e.g. the glutamine concentration must be higher than 1 mmol/l. It was convenient to use glutamine as a control-substrate for regulation of the medium feed rate. The dependency of product titer and space time yield from medium feed rate and substrate concentrations were studied.
