Experience with new assays for oestrogen receptors using monoclonal antibodies

Popis: The oestrogen receptor ( E R ) is a unique oestrogen-binding protein. (M, 66 182) [I]. found in the target cells of oestrogen action. The presence of E R in breast cancers indicates a better prognosis and greater likelihood of response to endocrine therapy [2]. The preparation of monoclonal antibodies against the human ER [3] has lead to the development of new histochemical (ER-ICA) and biochemical (ER-EIA) methods for detecting the protein. We have compared these new assays with our standard radioligand-binding assay [4] (DCC), established 13 years ago, and have related the results for the ER-ICA to response to endocrine therapy. Breast cancers ( I56 in total) were collected from patients undergoing mastectomy or biopsy, and used either fresh or alter storage in liquid nitrogen. For 41 patients, tumour cells were also taken by fine-needle aspiration a t initial presentation; these aspirates were treated with water to lyse erythrocytes and the malignant cells were collected on t o slides by cytocentrifugation according to R. E. Leake, (personal communication) before fixation according to the manufacturer's instructions (Abbott Laboratories Ltd, The Business Centre, Molly Millers Lane, Wokingham. Berkshire). Nine
