The estimation of the relative concentration of pure constituents in biochemical mixtures using their respective pyrolysis—mass spectra may be achieved by regression or principal components analysis. The duality of the two methods is illustrated using simple Euclidean geometry. Principal component projection of pure spectra on to the mixture space is shown to be equivalent to target transformation. Both methods are applied to artificially constructed and real pyrolysis data of mixtures of glycogen, dextran, and bovine serine albumin. No systematic departure from the additive model for real mixtures was observed. Recommendations on the design and improvements in analysis of such mixture studies are made. |
