EFFECT OF GABA ON SPONTANEOUS AND EVOKED RELEASE OF 3H-NA FROM SYNAPTOSOMES OF RAT BRAIN MESODIENCEPHALIC REGION

Popis: This chapter discusses a study to examine the effect of GABA on spontaneous and evoked release of 3 H-NA from synaptosomes of rat brain mesodiencephalic region. GABA in concentration 10 −4 and 10 –3 M has been shown to increase the spontaneous release of H-NA from synaptosomee of mesodlencephallc region. This effect appeared immediately after the addition of GABA and lasted about 1 min. The effect of GABA was blocked by 3 × 10 −6 M and 10 −5 M bicuculline that itself stimulates the spontaneous efflux of 3 H-NA and this effect developed gradually. GABA significantly inhibited electrically evoked release of 3 H-NA from synaptosomes. In the presence of bicuculline, the effect of GABA was not altered, whereas bicuculline as a sole substrate reduced the electrically evoked release of 3 H-NA. GABA blocked the K + -evoked (40 mM) and protoveratrine-evoked (5 μM) release of 3 H-NA from synaptosomes. In the Ca-free medium, the effect of GABA on the K + -evoked release of 3 H-NA was reversed. The results obtained suggest the role of GABA in the presynaptic regulation of NA release in brain.
