CD36 Expression in Human Monocytic Leukemia Cell Lines: THP-1 and THP-1 Subtype, Show Different Expressions of Type I and Type II Scavenger Receptors

Popis: We investigate the expression of CD36 in THP-1 cells and the THP-1 subtype (sTHP-1). The sTHP-1 cells accumulated esterified cholesterol after incubation with oxidized LDL (Ox-LDL), but not after incubation with acetylated LDL (Ac-LDL). The specific binding, association, and degradation of Ox-LDL through CD36 were 36, 134, and 179 ng/mg protein in parent cells, and 40, 111, and 174 ng/mg protein in sTHP-1 cells, respectively. Flow cytometric analysis showed that these cells expressed similar amounts of CD36 on their cell surfaces. We conclude that these cells expressed similar amounts of CD36, and that TGF-βl was likely to have little effect on the expression of CD36, at least in these cell lines. While type I and type II scavenger receptors bound and internalized both Ac-LDL and Ox-LDL, CD36 bound and internalized only the latter Ox-LDL.
