The diagnosis of Q fever is based on the detection of serum antibodies to Coxiella burnetii antigens. The isolation of the aetiologic agent is not recommended as a routine procedure due to difficulties in growing these bacteria and their infectivity. Since it has been well documented that infected animals and humans excrete bacteria in urine and semen, we have developed a dot-ELISA test and indirect immunofluorescence assay (IFA) for diagnosing the infection. The minimum concentration of bacteria detectable in urine with dot-ELISA was estimated as 1 × 10 6 cells ml −1 . IFA was less useful because single C. burnetii cells are hardly visible. In semen C. burnetii bacteria were detected equally well with both IFA and dot-ELISA test. |
