Publisher Summary This chapter provides an overview of harvesting human leukocyte cultures for chromosome cytology. The methodology for determining the chromosomal constitution from a sample of dividing leukocytes obtained from blood is relatively simple but the profusion of published procedures which differ in certain small details confuses or puzzles the investigator beginning such work. Although many substitutions can be made within a proved procedure, only with experience can one safely judge which detail may be modified. No single factor contributes more to the success of any metaphase chromosome technique. In scanning a preparation one can visually reject in a few seconds dozens of poorly spread or poorly fixed metaphase cells and examine those few which happen to prove suitable. Even with optimal technique, when growth has been poor, the results will be correspondingly poor or so sparse in metaphase cells of suitable quality that much time would be wasted on such material. The necessity of selecting the better metaphase cells for chromosome cytology studies makes valid comparisons of minor changes in technique difficult. |
