The method Magnetically Induced Chemical Analysis (MICA) has been developed for the automation of affinity assays. The method is based on manipulation of paramagnetic, spherical particles in stationary liquids. The particles are coated with affinity ligands, which allow both for selection and preconcentration of analyte. The interaction between the reagent on a particle and the analyte in a sample has been simulated. The accumulation of analyte on the particle appeared to be strongly dependent of its analyte binding capacity. For higher binding capacities analyte depletion occurred around the particle and in that case the particle movement created in MICA would increase the rate of preconcentration. Besides higher preconcentration rates the particle with high binding capacity would be sensitive to the analyte in a wider concentration range. |
