A NOVEL MAMMALIAN EXPRESSION SYSTEM BASED ON THE GLOBIN LOCUS CONTROL REGION

Popis: We have developed vectors using the globin locus control region (LCR) that support high level expression of heterologous proteins in MEL (murine erythroleukaemic) cells when induced to differentiate with dimethylsulphoxide (DMSO). The inclusion of the human beta-globin promoter and the 3′ prime untranslated sequences of the human beta-globin gene in the vector allows high level expression. Accumulated product levels greater than 100 mgl-1 have been obtained. Passage of clonal populations with and without selection pressure for greater than 40 generations has shown no reduction in copy number or productivity. MEL cells exhibit erythroid characteristics and it is therefore possible that they may have poor secretory capacity. However, we shot that induced MEL cells are capable of efficiently secreting heterologous protein via the golgi/endoplasmic reticulum.
