In the last decade, a growing body of evidence has further challenged the principle that proteins required at different subcellular regions of neurons are exclusively synthesized by the soma. Experiments performed on dendrites and axons have suggested that targeting of mRNAs and their subsequent translation is a mechanism by which proteins are provided to various subcellular domains. However, direct experimental evidence for the existence of such a mechanism is still lacking. To demonstrate a direct relationship between the presence of a given mRNA and its product, we developed an experimental system consisting of identified neurons from the mollusc Lymnaea stagnalis. We injected a heterologous mRNA into isolated axons of the cultured identifiable neurons and showed the translocation of the mRNA into the neurites emanating from the isolated axon. Next, we demonstrated the translation of the injected mRNA by immunocytochemistry. The translated product appeared to be confined to the varicosities and growth cones. The involvement of protein synthesizing machinery was indicated by the identification of ribosomal RNA using both in situ hybridization and electron microscopy. |
