Recombinant animal cell lines for production of glycoproteins

Popis: Eukaryotic expression plasmids for constitutive or inducible expression with different transcription control and RNA processing elements were constructed. The plasmids contain human structural genes coding for Antithrombin III (ATIII), Interleukin-2 (IL-2) and Interferon-s (IFN - s). The plasmids were stably transferred into three different mammalian cell lines, hamster CHO, BHK21 and mouse L cells. The expression of the gene products reflects predominantly the activity of the transcriptional control elements in different cell lines and the influences of the respective structural gene on the posttranscriptional level. Selected transformants were screened for high expression by clonal selection before and after gene amplification. The productivity is up to 10 μg/106 cells/24 h. The products were characterized by biochemical, immunological and biological methods. In all cases biological activities are indistinguishable from the authentic gene products. Analysis of the carbohydrate moieties indicates that composition and extent of glycosylation are cell type specific.
