Publisher Summary This chapter describes the basic methods for investigating the biosynthesis of sodium channels, and outlines the use of these procedures to deduce certain characteristics of these complex molecules. Sodium channels purified from a number of tissues show significant nonprotein domains. Because the sodium channel is heavily posttranslationally modified, these methods focus on the biosynthetic events occurring in the rough endoplasmic reticulum (RER) and Golgi apparatus as the channel molecule is processed from an initial immature core polypeptide to a mature lipoglycoprotein. The chapter presents an investigation of the biosynthesis of electroplax sodium channels both in isolated cells from the electric organ of the eel and in Xenopus oocytes injected with electroplax mRNA. Evidence of posttranslational modification may be obtained by comparing the physicochemical characteristics of the mature protein as isolated from the plasma membrane with the unmodified protein core. With appropriate means of detection, these methods should be readily adaptable for similar studies on other ion channels. |
