Current understanding of transposable element activity is restricted due to limited study methods. Sequence analysis or bulk measurements fail when information is lost in ensemble averages or affected lineages go extinct and do not remain in the phylogenetic record. We have developed a method for studying the activity of the transposable element IS608 in Escherichia coli using real-time detection techniques combined with microfluidic platforms, providing an opportunity to study natural IS608's activity in vivo, with single cell resolution and real-time tracking. IS608 was originally found in Helicobacter pylori. Here we describe our quantitative measurements of IS608's activity in Escherichia coli and discuss their implications. |
