Recently we and other investigators have used the reaction of light with the photosensitizer Rose Bengal (RB) as a tool to investigate the effects of reactive oxygen species (ROS) on cardiac tissue. These studies have provided new and useful information regarding ROS effects on the action potential and ionic currents in cardiac tissue. The favorable comparison (see below) of the cardiac electrophysiological modifications produced by photoactivation of RB to those produced by other ROS generators supports the conclusion that photosensitizers provide a biologically relevant method to evaluate ROS-induced cellular damage. What follows is a summary of the electrophysiological modifications observed by us and other investigators studying the effects that photoactivation of RB has on whole heart and isolated cardiac cells. These results are compared to those that others have obtained using different methods to generate ROS such as (a) dihydroxyfumaric acid (DHF), (b) xanthine oxidase (XO), (c) hydrogen peroxide (H2O2), and (d) organic hydroperoxides such as tert-butyl hydroperoxide (TBH) and cumene hydroperoxide (CH). The relevance of these studies to electrophysiological modifications in ischemic-reperfusion injury is also presented. |
