Popis: | AY-9944 [trans-1,4-bis(2-chlorobenzylaminomethyl) cyclohexane dihydroohloride], an inhibitor of cholesterol biosynthesis, was studied for its effect on adrenal corticoid synthesis. In rat adrenal homogenates AY-9944 inhibited the production of corticosterone from cholesterol, progesterone and 11-desoxy corticosterone, but did not affect its formation from 11β-hydroxyprogesterone. Adrenal homogenates from AY-9944 treated rats showed little change in their 11β-hydroxylation activity of DOC. In rats, chronic treatment with high doses of AY-9944 depressed adrenal vein corticosterone levels and impaired the capacity of the adrenals to convert circulating cholesterol-7α-3H into corticosterone.
