Popis: | Soon after the isolation from beef parathyroids of an active principle, capable of raising the levels of calcium in the serum of normal and parathyroprivic dogs (1), Collip (2) and others (3, 4, 5, 6) found that repeated, moderately large doses of the extract were extremely toxic. They observed that the injection into dogs of about 125 I.U. of parathyroid extract at frequent intervals produced a fairly rapid rise in the levels of serum calcium, which reached a concentration of about 20 mg. per cent in 20 hours and remained at this level for some time. However, in spite of the administration of the extract, the serum calcium concentration usually declined, and the inorganic phosphorus increased, in about 36 hours. The increased excretion of calcium and phosphorus in the urine, which began shortly after the injections were started, then declined sharply as though the injections had been interrupted. The concentrations of urea and of nonprotein nitrogen in the blood increased steadily, and were followed late... |
