The stability of interleukin-2 mRNA transcribed in splenic CD4+ T cells from SAMP1 mice

Popis: SAMP1 mice show markedly low T-dependent antibody-producing responses. Recently, we demonstrated that splenic CD4 + T cells from SAMP1 mice show abnormally short-lasting production of interleukin (IL)-2, leading to the impaired proliferation and survival of these cells. Therefore, we examined the expression of IL-2 mRNA and its stability in the SAMP1 CD4 + T cells stimulated with Con A by RT-PCR and actinomycin D treatment, and compared expression with that in cells from C3H/He. The CD4 + T cells from SAMP1 showed a sudden decrease in the amount of IL-2 mRNA 15 h after stimulation, though the RNA was detected even at 24 h in C3H/He. Moreover, the rate of IL-2 mRNA degradation under transcription block was faster in SAMP1 than in C3H/He cells. These results suggest that the increased degradation of IL-2 mRNA may be the cause of insufficient IL-2 production by the SAMP1 CD4 + T cells.
