WNT5A and CREB genetic profiling from breast cancer patients: A case study of Malang population

Popis: Indonesia has the highest prevalence of breast cancer in Southeast Asia, and East Java as the province with the most cases. Previous research confirms 10% of breast cancer cases are caused by genetic factors. Various studies on the molecular mechanism of breast cancer are carried out to find new targets for research and therapy. WNT5A is one of the genes reported has a function in the development of cancer, but on the other hand, it showed tumor suppressor function. WNT5A is reported to play a role in CREB activation, a transcription factor that confirms its upregulation plays a significant role in breast cancer metastasis to bone. This study aims to determine the expression level of WNT5A and CREB in breast cancer samples from patients in East Java with a known stage. WNT5A and CREB expression levels were examined through RT-qPCR. The results showed no similar pattern for both WNT5A or CREB expression with an increase in breast cancer stage. This is suspected because both WNT5A and CREB have a dual role in breast cancer. The role of WNT5A is determined by the isoforms it produces, WNT5A-long (WNT5A-L) and WNT5A-short (WNT5A-S). While the role of CREB is determined by its activation pathway, another study needs to be done by examining the expression of WNT5A isoforms and other genes that play a role in CREB activation.
