Src and Src family members are non-receptor protein-tyrosine kinases that play key roles in cellular function. v-Src (a viral protein) is encoded by the chicken oncogene of Rous sarcoma virus and Src (the cellular homolog) is encoded by a physiological gene, the first known proto-oncogene. Src contains SH3 (Src homology domain 3), SH2, and SH1 (protein kinase) domains. These domains were among the first to be characterized as protein–protein interaction modules. The regulation of Src is intricate and involves protein kinases and phosphatases as well as transmembrane receptor protein-tyrosine kinases, G-protein coupled receptors, and integrin, cadherin, and immune receptors. |
