The liver of a human accidental poisoning victim who injested the rodenticide Vacor was examined for residues of the active ingredient 1-(3-pyridylmethyl)-3-(4-nitrophenyl) urea (RH787). A trace analytical method for RH787 using the polarographic reduction of the nitro function was developed using differential pulse polarography (DPP) which permitted detection of 2 × 10 −8 M RH787 (17 ng RH787/3 ml). The nitroaromatic metabolite identified in the sample was p-nitroaniline (PNA), which has polarographic behavior similar to that of RH787. Peak potentials in DPP were: unknown, −571 vM vs SCE; RH787, −585 mV; PNA, −570 mV). Thin layer chromatographic R f values for the metabolite corresponded to those for PNA. The identification was confirmed by gas-liquid partition chromatography on 5% OV-210 and 1.5% OV-17/1.5% OV-210 columns. The presence of PNA at 5.1 ± 1.0 mg PNA/kg liver suggests amide hydrolysis of RH787 . |
