The effect disease states can have on drug metabolism and disposition of prescribed and over-the-counter drugs is not well understood. The absorption, distribution, metabolism, and elimination (ADME) of drugs can be influenced by the physiological and biochemical changes that occur due to various diseases. These changes can have dramatic influence on the exposure of drugs and lead to drug toxicity or lack of drug efficacy. Herein we describe the general principles of drug metabolism and how various disease states can affect the ADME properties of drugs. |
