Full-length genome analysis of an avian influenza A virus (H9N2) from a human infection in Changsha City

Popis: | Aim: Sequencing and phylogenetic analysis of avian influenza A (H9N2) virus strain, which was isolated from a 2-year-old child presenting influenza-like symptoms. Methods: Viral genome was acquired by RT-PCR, and phylogenetic trees were constructed using the neighbor-joining method. Results: A/Hunan/44557/2015 sequence shared identity with the RSSRGLF motif – first reported in low-pathogenic avian influenza in birds. Polymerase L336M and hemagglutinin Q226L mutation was found in the strain. Two newly mutation sites, T197D and D483N in hemagglutinin gene, were found. Phylogenetic tree analysis revealed that the eight gene segments of this virus contained three lineages. Conclusion: A/Hunan/44557/2015 may have acquired the ability to infect humans via genetic exchange with other H9N2 viruses, indicating that the H9N2 genome can generate pandemic isolates. |
