Popis: Percoli and sucrose gradients were used to isolate respectively heterocysts and vegetative cells of Anabaena azollae symbiont of Azolla filiculoides. Both heterocysts and vegetative cells devoid of contaminating bacteria and Azolla debris, were used to assay the presence of the enzyme Superoxide dismutase (SOD) by activity-stained polyacrylamide gel electrophoresis. Types of SOD were determined by different sensitivity to 2mM cyanide and 2mM hydrogen peroxide. Heterocysts contained only a faint band of both hybrid iron-manganese (Hy) and iron (Fe) Superoxide dismutase. Vegetative cells possess Mn-SOD, Hy-SOD and Fe-SOD. Hy-SOD is detected for the first time in cyanobacteria. The higher activity was present as Fe-SOD in vegetative cells while total heterocysts SOD activity was only 15% with respect to vegetative cells.
