The effects of linoleic acid feeding on renal growth and function in uninephrectomized rats

Popis: Pharmacological inhibition of cyclooxygenase limits renal growth and function following uninephrectomy in rats. Chronic linoleic acid feeding augments renal prostaglandin and thromboxane synthesis in partially nephrectomized rats and slows the pace of development of glomerulosclerosis. The present study was undertaken to evaluate the effects of linoleic acid feeding on the acute adaptive changes in renal mass and function which follow uninephrectomy in rats. Young adult male Sprague Dawley rats were fed a diet rich in linoleic acid or pair-fed a control beef tallow-rich diet. Two weeks after initiation of the diet, the animals underwent uninephrectomy. Two weeks later, urine was collected for prostaglandin E 2 excretion and the clearances of inulin and PAH were measured. Linoleic acid feeding enhanced excretion of prostaglandin E 2 , and dramatically reduced the clearances of inulin and PAH. Despite marked diminution in renal function, linoleic acid-fed rats had an identical renal growth response to beef tallow-fed controls. These data provide novel evidence against the "work hypertrophy" theory of renal growth, and support an important role for prostaglandins as mediators of both the functional and anatomical responses to a reduction in renal mass.
