Decreased dihydromorphine and D-ala2-D-leu5-enkephalin binding after dynorphin pretreatment in mice

Popis: Dynorphin(1–13) has been shown to antagonize morphineinduced analgesia and to inhibit opiate receptor binding in vitro . In this study, we pretreated mice with dynorphin; opiate receptor binding was then performed in crude P 2 fractions in vitro , using radioactive dihydromorphine and D-ala 2 -D-leu 5 -enkephalin. It was found that both dihydromorphine and D-ala 2 -D-leu 5 -enkephalin binding were significantly decreased by in vivo dynorphin treatment. Dynorphin's effect was both dose- and time-dependent; maximum effects were obtained at 30', lasted up to 3 hrs and were fully recovered by 12 hrs.
