CLONING OF ENTEROBACTER CLOACAE CEPHALOSPORINASE GENE AND ITS EXPRESSION IN ESCHERICHIA COLI

Popis: The chromosomal gene for cephalosporinase (CSase) production by Enterobacter cloacae GN7471 was cloned into vector plasmid pACYC 184. From the substrate profile and the response to anti-E. cloacae CSase serum, it was shown that the hybrid plasmid (pMS161) carries the chromosomal CSase gene of E. cloacae GN7471. E. cloacae CSase encoded by the cloned plasmid was produced in an inducible manner in both E. cloacae and E. coli, but its expressed specific activity was much lower than that of the original E. cloacae. From the analysis of several hybrid plasmids derived from pMS161, it was deduced that the site of the structural gene of CSase is located in a 1.15-Md fragment and the gene regulating the inducibility of CSase prodution in a 0.3 Md fragment.
