A method for the isolation of plasma membranes from rat uterus and rat duodenum

Popis: Summary For studies of binding the peptide hormone bradykinin to its specific receptor a method has been developed for isolation of plasma membrane fractions by the same procedure from both rat uterus and rat duodenum.The yield of plasma membranes reached 1% as calculated from the initial protein content of the homogenate.The plasma membrane fraction has been characterized by estimation of the activity of some marker enzymes, especially of 5'-nucleotidase (EC 3.1.3.5) and of ouabain-sensitive (Na + , K + )-ATPase (EC 3.6.1.3) which has been demonstrated to exhibit an increased activity in the plasma membrane fractions.The content of both cholesterol and phospholipids are highly enriched.The membrane fractions could be stored in the cold for several weeks without loss of activity.These membrane fractions have been successfully used for binding studies of brady-kinin and for demonstration of enzymatic degradation of this hormone.
