Impaired arachidonic acid metabolism of platelets and vessel wall in experimental diabetic rats and its restoration by insulin administration

Popis: | It has been generally accepted that the loss of balance between vascular prostacyclin (PGI2) and thromboxane A2 (TXA2) takes part in the development of the vascular injury on the diabetic state. This paper reports the effects of insulin administration on the disorders of prostaglandin and on lipid metabolism in the streptozotocin (STZ) induced diabetic rats. For the control of the diabetic state, lente insulin (50-100U/kg) was administered subcutaneously until the achivement of normal blood glucose level in the diabetic rats. After 9 days, plasma glucose and lipids were measured by conventional methods, and both TXA2 generation in the platelets and PGI2 generation in the isolated aorta were measured by RIA.The results were as follows: 1) plasma glucose, total cholesterol, atherogenic index, triglyceride, free fatty acid and β-lipoprotein increased in the diabetic rats, 2) Vascular PGI2 release decreased significantly, while platelet TXA2 generation induced by ADP, collagen, thrombin and arachidonic acid increased in the diabetic rats, 3) The administration of insulin corrected the elevated lipid level, 4) The decreased vascular PGI2 generation was restored and the increased platelet TXA2 was reduced by the administration of insulin.From these results, the impaired arachidonic acid metabolism of platelets and vessel wall in the diabetic rats was considered to be corrected by the treatment with insulin. |
