DEVELOPMENT OF SERUM FREE MEDIA FOR ENGINEERED NSO CELL LINES

Popis: A serum free medium supporting growth and antibody production in a recombinant NS0 cell line was developed. Initial experiments focused on a formulation which would support growth and antibody production for one passage in serum free medium in T-flasks. A number of commercially available media were screened but they did not support cell growth, either when cells were weaned into serum free medium or inoculated directly into serum free medium. Cells were then grown in an in-house serum free medium, YD25, based on supplemented William's Medium E. Further supplementation of this medium with different lipid cocktails was found to give growth and antibody production similar to that in serum-containing media in T-flasks. Several of these media formulations were tested for their ability to allow long-term propagation of cells, and the growth rates and antibody production in these media are shown.
