Popis: | Lac-lux marked Pseudomonas aeruginosa UG2Lr cells were encapsulated in alginate or a novel κ -carrageenan formulation. The beads were dried and maintained at 4°C in the dark. Plate counts determined that survival of encapsulated cells, after an initial Log 2 decrease over 24 h to Log 7.5 CFU g −1 dry beads during drying, only slightly decreased to Log 6.8 CFU g −1 dry beads after four years in storage. The lux activity of the genetically-modified bacteria was determined using a Biomedical Image Quantifier and all resulting colonies expressed luminescent activity. These results demonstrate the potential for long-term storage and subsequent use of encapsulated bacterial cells for environmental, as well as other applications. |
