A variety of oxidants generates nucleic acid modifications (lesions) via hydrogen atom abstraction from the carbohydrate backbone. In addition, nucleobase radicals are a hallmark of ionizing radiation (γ-radiolysis). The initially formed carbohydrate and nucleobase radicals, as well as the resulting peroxyl radicals, can react with proximal nucleotides to form tandem and other clustered lesions. Clustered lesions can be more difficult to repair and/or bypass than isolated lesions. Consequently, they are potential biological threats. |
