Prostate cancer bone metastasis, occurring in the advanced stages of cancer, involves intricate and complex processes which need to be simplified in order to alleviate the plights associated with the disease. The ability of bone to facilitate prostate cancer metastasis is an important aspect of the disease which needs to be understood better. 3-D in vitro models present a great platform to conduct studies to elucidate the multiple facets of prostate cancer within the primary tissue and also, in the secondary bone tissue. The ease of formulating and fabricating a 3-D microenvironment is beneficial in understanding the role of various components of extracellular matrix in the development and progression of prostate cancer. The versatility of this medium to not only investigate the molecular basis of the prostate cancer activity within the bone metastatic site but also finds its application in revealing potential therapeutic targets and testing treatment efficacies. This review is a consolidation of the tissue-engineered 3-D prostate cancer models along with its scope and future directions. This review also discusses the development of bone metastatic prostate cancer and the various strategies adopted by the cancer cells to ensure successful colonisation at the secondary site. |
