Two nuclear localization signals required for the nuclear localization of rice ribosomal protein S4

Popis: Several chromosomal proteins have been isolated to date possessing ability to translocate from cytoplasm into nuclei in eukaryotic cells. A gene encoding rice small subunit ribosomal protein S4 was previously isolated and two putative nuclear localization signals (NLSs) were found at each end of S4 protein. In this report, by using fusion protein with green fluorescent protein (GFP), we demonstrated that S4 protein is localized to the nucleus of onion epidermal cells. The two putative NLSs were analyzed for a possible role in the nuclear localization of S4 protein. It was found that both of the two NLS, regions from amino acids 5 to 11 and 239 to 255, are responsible for nuclear localization of the S4 protein. Deletion in either part of these regions caused significant loss of nuclear localization pattern of S4 protein. Moreover, coordination of the two regions may be important for the S4 nuclear localization, due to the fact that substitution of several conservative residues did not effectively affect the nuclear targeting.
