Popis: The mouse has long contributed to our understanding of mammalian physiology and disease. Owing to its short life cycle, fecundity, and amenability to genetic manipulation, the mouse is a powerful tool for modeling human disease. This chapter discusses the basic methodology used in the creation of mouse models and illustrates their strengths and limitations. Ongoing challenges include modeling complex multigenic disorders, and validating genetically engineered mouse models in drug development and preclinical testing. Fortunately, the advent of rapid, low-cost in vivo somatic modeling techniques (e.g. CRISPR), enhances the utility and impact of mouse models in biomedical research.
