Effects of Growth Factors on Hybridoma Culture in the Perfusion System

Popis: From the experiments using a perfusion culture system with a cell sedimentation unit and a separation unit composed of a hollow-fiber module, the effects of. growth factors such as transferrin and BSA were analyzed. Transferrin enhanced the cell growth but had no significant effects on the specific antibody production rate. BSA promoted cell growth and raised the specific antibody production rate. When the fresh medium containing 2 g/l. or 5 g/l of BSA was fed into the flask, both the specific growth rate and specific death rate increased. The specific antibody production rate was increased 2 times and 25 times by feeding 2 g/l and 5 g/l. of BSA, respectively, compared with no addition of BSA. In the continuous culture without the separation membrane, BSA addition had no effects on the antibody production.
