The association of washed ribosomal subparticles from Bacillus stearothermophilus was carried out by incubation with the association factor. Several properties of the resulting 30 S–50 S couples were compared with those of the “native” 70-S ribosomes isolated from bacterial extracts after slow cooling of cultures. The following observations were made: 1. 1. 70-S ribosomes were more resistant to the action of the dissociation factor, pancreatic ribonuclease and the hydrostatic pressure generated during sucrose gradient centrifugation at high speed. 2. 2. The subunit exchange with radioactive 30-S particles was lower for 70-S monomers than for associated subunits. 3. 3. Both particles had identical activities in the polypeptide synthesis reaction when they were incubated in the presence of a random poly(U,C) as messenger. |
