Peptides YYGGEGSSSEQG and SESEM Inhibit TNF-α-Induced Smooth Muscle Cells Proliferation and Migration Through Their Bindings to TNF-α Receptor

Popis: The peptides YYGGEGSSSEQG and SESEM derived from rice α-globulin have been reported to possess anti-atherosclerotic activity, but the role in the proliferation and migration of vascular smooth muscle cell (VSMC) and the underlying mechanisms remain unknown. In the present study, we observed the effects of the peptides YYGGEGSSSEQG and SESEM on VSMC proliferation and migration, and a computer simulated interaction between the peptides and TNF-α. The peptides might effectively inhibit TNF-α-induced VSMC proliferation and migration or promote its apoptosis by promoting the expression of transforming growth factor-β (TGF-β) and suppressing the expression of platelet-derived growth factor (PDGF) BB proteins. In addition, the binding energy of YYGGEGSSSEQG to TNF-α receptor 1 (TNFR1) was lower than that of endogenous RMP16 with high activity. Therefore, the peptide inhibited the activity of TNF-α by binding to TNFR1. These results demonstrated that the peptides YYGGEGSSSEQG and SESEM reduced TNF-α-induced smooth muscle cell proliferation and migration or increased its apoptosis by promoting the activity of TGF-β and inhibiting the activities of PDGF-BB and TNF-α.
